The film, set after the events of The Amazing Spider-Man 2, where Peter has found new friends in Mary Jane Watson (Shailene Woodley) and Eddie Brock (Finn Wittrock), but everything changes with Oscorp employee Felicia Hardy (Felicity Jones) becoming the vigilante Black Cat, and the return of Harry Osborn (Dane DeHaan), Peter's friend turned enemy. Spider-Man sees that the only way to stop Ultimate Green Goblin will end up putting his life at risk, since brute force alone had been proven ineffective. Spider-Man fights Mysterio.

Atop Lower Manhattan's upper walkway, Spider-Man had Mysterio captured until Mysterio called on E.D.I.T.H. to summon drones to attack Spider-Man. As he narrowly dodged the drones, Spider-Man ended up at the opposite end of the walkway from Mysterio. Taunting the young hero to take the glasses from him, Mysterio activated illusion projectors to create a void of darkness. To counter, Spider-Man calmed himself and closed his eyes, having chosen to rely on his "Peter Tingle" to navigate.

Spider-Man charged into the walkway blind, fully cognizant of the drones hidden by cloaking, and proceeded to destroy them which ended the darkness illusion. Spider-Man closed the distance between him and Mysterio, but in raging desperation, Mysterio commanded E.D.I.T.H. to order his orbiting defense drones to fire on Spider-Man. This tactic failed, as the young hero used one drone as a shield to break another drone, and the second to last drone to fall misfired and hit Mysterio in the abdomen. Peter's nuclear power begins to manifest itself, but this time he is unable to control it.
